It almost looks like it. I think it’s just a lighter patch of grain.

Strange how with my 3x mag lighted goggles its almost harder to pickup, but it sure looks circular and matching both sides of the headstock. If it is a dowel it looks like a pretty good job. I'm weary with current Texas weather drama and a previous bad experience, I may just keep it since its going on my Uglycaster. Allparts usually need some filling with CA at some fret ends, this one is fairly uniform. Fretboard is straight, haven't got out the fret rocker. Pretty doggone inexpensive, nice soft vee profile. I have a message in to the seller asking if this is a known "b stock".

The edges don’t seem “perfect” enough to be a dowel. At least from what I can see online.

You can fix that. Just drill it out a little bit bigger and plug it with a large dowel. That light spot (whatever it is) will be all gone.  |

A trusted friend says its hard to tell but based on the grain movement around the circular spot thinks maybe a branch, suggested dampening it to see if it reveals any raised woodgrain. I'm gonna chalk this one up to "I dunno" and move on with the build.

Nah, not a dowel. If you look at the first picture, you can see some grime in the very small grain pores, and the pattern holds across. Dowels will end up poking end gain out where long grain is expected and would be more noticeable.
One possibility is that it is a drip spot. Maybe a bit of solvent or water hit it at some point and it had a very slight discoloring effect. It might go away with a wipe of whatever or hitting lightly with a sanding or buffing pad. If Allparts puts any kind of sealer on there like Warmoth does, it would make it all the more visible.

I've plugged many a hole in maple. First, you would never use a dowel unless you wanna make sure it's as obvious as possible; you use a plug cutter so you can get similar grain orientation. To get one to disappear completely is pretty much impossible though if you get to resurface the area afterward it will generally bu unnoticeable to the casual glance. I just yesterday plugged one in a finished maple kitchen counter support I built. These situations (a hand-drilled hole with forstner bit, fluch trimmed, scraped and finished) will usually be pretty obvious as you just can't get a perfect hole when drilling by hand.
All that said, yours doesn't look like a plug to me. You'd be able to see the outline upon close inspection and you can see color variations spanning what would be the edge of the circle. Not impossible but very very unlikely.
