Considering I have more pedals than most guitar stores...it's hard to narrow it down.
The essential for me is a Space Echo preamp section (even if the echo is off) at the end of the chain punching the amp in the face.
Past that, I have a double pedal made to my specification with switchable order (so I don't have to run cables, just flip a switch): a Roland Bee Baa boost section (it's very dirty and punchy) feeding a germanium Rangemaster clone with a bit more punch on tap and adjustable bias, or vice versa.
For my non-live room rig, I've been very happy with a TC Electronics Klon clone doing a similar function into the recording console - where I use a Pultec tube EQ clone as a channel strip feeding the amps and the rack of gera.

Being more of a bass player than a guitar player these days, my pedal array is pretty simple: When I do play guitar, it's just a 335 loaded with Humbucker-sized P-90-esque pups straight into a 5E3 Tweed Deluxe clone (Clark Beaufort Special).

I'm a simple guy with simple tastes. Usually I roll with a Ibanez tube screamer (or a fuzz) and work the volume knob for texture. I use a TS808Ri.
I stack a TS808Ri behind a TD9DX. I step on the TS9DX for solos and any rock applications.
One day it dawned on me that Ibanez must have designed that TS9DX to stack with a Tube Screamer. The TS9DX is a stock TS9 with 3 other settings that progressively get lower in gain,(i.e cleaner) but more pronounced in base, headroom and signal gain.
I use the setting up from the stock TS9. Excellent tone dynamics, works really well with all my amps. (anything one thinks a normal tube screamer lacks, with one of those channels... its all there)

I have two TS9DXs. This means I don't have to move any pedals to stack one ahead of the other. Pretty handy, right ? And no, I'm not being serious about the above, though I do have two TS9DXs. You might find more plentiful tech info on tdpri. I think there's a whole section for simple people who stick their fingers and soldering irons in deadly tube amps and then want to torture the poor things with distortion stacks ! My serious stack rig is a Plexi Van Gogh into a Da Vinci Mona II into a Picasso2000 into a delay and into a Pignose 7-100. What ? That's artistic melting lead tone right there !

Well, for the last month or so, with my Marshall Origin 50, I'm liking the Soul Food into a Plimsoul. That offers me various flavors combined with my volume knob, and gain boost switch on the amp. Great with a Les Paul and even makes my Strat sound fat.

For non-overdrive, well, the standard issue my entire playing career damned near has been a Roland Space Echo of some type (the preamp alone should count as an effect, bujt....I get off cheap here!) and when I had a choice or the budget, two of them stacked.
I can do three now, one original tape unit and two RE-20s. :0
